This study conceptually redefines rocaglates as translation remodelers that reprogram both the translation machinery, e.g., increasing eEF1ε1 translational activity, as well as the translatome, including the upregulation of numerous proteins, e.g., GEF-H1, that mediate rocaglate cytotoxicity in cancer cells. As a proof of concept, GEF-H1 activates RHOA, leading to increased JNK phosphorylation and apoptotic signaling resulting in reduced cell viability.
